Morphology is the assessment of the size, shape, and structure of individual sperm cells. In the cattle industry, it is a critical component of the Bull Breeding Soundness Evaluation (BBSE), as it is the trait most strongly correlated with a bull’s calf output.

A normal bull sperm cell consists of three main parts:

  • Head: Contains the highly condensed genetic material (nucleus) and the acrosome, which has enzymes necessary to penetrate the egg.
  • Midpiece: The “engine room” containing mitochondria that provide energy for movement.
  • Tail (Principal Piece): Provides the locomotion required to reach the egg.

Defects are typically categorised based on their origin or their impact on fertility:

  • Primary Defects: Occur during sperm production (spermatogenesis) in the testes. These usually involve the sperm head (e.g., pear-shaped, narrow, or cratered heads).
  • Secondary Defects: Occur after the sperm has left the testes, during maturation in the epididymis. These often involve the tail or midpiece (e.g., bent tails, cytoplasmic droplets).

  • Gold Standard: Professional assessment typically uses Differential Interference Contrast (DIC) microscopy at 1000x magnification to see subtle defects.
  • Passing Threshold: A bull is generally considered to have satisfactory morphology if at least 70% of its sperm are morphologically normal.

  • Genetics: Certain breeds (e.g., Brahman vs. Angus) have different baseline morphology profiles and predispositions to specific defects.
